I would not build anything over a 3.750" stroke X 4.185" bore without using an aftermarket tall deck block , and nothing with a crank and rotating assembly that I was too fond of if your looking at producing a lot of power .

the biggest cubes that i know of, coming out of a standard GM small block is 421 from a 400 but you need to grind alot from inside the block (pan rails and bottom of cylinder walls).
we run a 415 (3.8 stroke) from a gm 400 block. had to do the same work (grinding for rod clearance) and i'm not convinced it was worth the effort.
myself, i'd look at 383 next time or 372.
