If I run a bypass fuel relief valve on a return line from the back of my fuel log do I have to have a vent spot on the fuel cell also? What I mean is do I have to have a pickup line, return line, and vent line? Or will a pickup and return line work right?

Just use a vented cap.

yes, you will have to run a vent of some sort. personally i'm not a fan of the vented caps but thats just me.
you have to run a vent because the fuel being pulled from the tank needs to be replaced with air (or the tank will collapse).
